Winning the toss and batting first, Will Mason was bowled by Josh Smith for seven and Smith also bowled George Gray to make it 25-2.
Overseas player Callum Hotham, playing his last game for the club, and skipper Harry Ellison put on some good runs together before Ellison was caught by Reece Croker off Chris Day, having added a useful 30.
Ali Easton added 15 before being run out with Glastonbury on 110-4.
Hotham reached his 50 with a four and added eight more runs when he was out bowled by Day.
John Stratton continued to bat confidently, hitting ten fours and three sixes on his way to 91.
The final wickets fell cheaply as Glastonbury were all out for 252.
The hosts struck early to dismiss Lansdown's Jordan Smith when he was caught by Ali Easton off Dan Williams' bowling.
The visitors had progressed to 57-2 when Steve Keates dismissed opener Joe Jenkins for 21.
Tom Hankins then built a strong partnership with Ngoni Mupamba until George Gray took a catch off Joe Mason's bowling to take the breakthrough wicket of Hankins for 64.
Mupamba (68) then batted with Reece Croker and also got his 50 before Williams struck again, making it 229-4.
Economical bowling from Mason and tight fielding, including a Stratton run-out, kept the run rate down before Mason took another wicket.
In a gripping finish, it fell to Williams to bowl the last over and Lansdown were on 250, needing three to win off the last ball.
